The Lo-D MT-150 is a water-soluble, methylene blue based dye primarily used for taxol hydrogen bonding assays. It can also be applied for measuring thrombin formation on surface plasmon resonance sensors and as a simple visual monitoring of enzymatic activity. This dye has a higher quantum yield than native methylene blue, making it more effective for use in these assays. Its water-solubility ensures that it can be easily dissolved and diluted for use in a variety of experimental conditions. The MT-150 formulation was developed as a replacement for the previous Lo-D MT-110 dye, offering improved light absorption and stability over a wider pH range.In taxol hydrogen bonding assays, the Lo-D MT-150 is used as a simple, visual monitoring tool to quantify the hydrogen bond formation between taxol and binding partners. The MT-150 dye molecules are labeled onto one component in the assay, allowing it to be readily distinguished from any other components. If taxol forms hydrogen bonds with the labeled component, it causes a significant change in the light absorption properties of the MT-150 dye, resulting in a measurable shift in absorbance at 652 nm. This shift can be used to quantify the proportion of labeled component bound to taxol, providing an accurate assessment of the assay's outcome.In addition, the Lo-D MT-150 can be used to monitor thrombin formation in a surface plasmon resonance (SPR) sensor assay. By attaching the MT-150 dye to one of the assay's components, the presence of thrombin can be detected as a result of uncoupling the dye from its original position. This uncoupling causes a decrease in light absorption, leading to a detectable change in the SPR sensor's angle-shift output, allowing for the quantification of thrombin concentration.Overall, the Lo-D MT-150 dye's versatility and improved properties make it a valuable reagent for numerous assay applications, such as taxol hydrogen bonding and thrombin assays.
